褪黑素对葡萄幼苗生长及硒富集的影响

Effects of Melatonin on Growth and Selenium Accumulation of Grape Seedlings

      目的  探讨在硒胁迫下外源褪黑素对葡萄生长和硒富集的影响,筛选出能够促进葡萄生长和硒富集的最佳褪黑素浓度。
      方法  以‘夏黑’葡萄扦插苗为材料,采用盆栽试验,研究了在硒胁迫下褪黑素对葡萄幼苗生长、生理及硒富集的影响。
      结果  不同浓度的褪黑素在一定程度上提高了葡萄幼苗的生物量和提高了根冠比,其中浓度为150 μmol L−1的褪黑素对葡萄幼苗地上部分生物量的提高幅度最大(18.22%)。褪黑素处理也在一定程度上增加了葡萄幼苗叶绿素和类胡萝卜素含量,提高了光合能力和抗氧化酶活性,降低了脯氨酸含量、可溶性蛋白含量、相对电导率和丙二醛含量。不同浓度褪黑素均提高了葡萄幼苗的硒含量,其中浓度为150 μmol L−1的褪黑素对葡萄幼苗地上部分硒含量的提高幅度最大(112.71%)。回归分析表明,褪黑素浓度与葡萄幼苗根系生物量、地上部分生物量、根系硒含量和地上部分硒含量均存在二次多项式或线性回归关系。
      结论  外源褪黑素能促进硒胁迫下的葡萄生长和硒富集,其中以150 μmol L−1的浓度为最佳。

     

    Abstract:
      Objective  The paper aims to investigate the effects of exogenous melatonin on the growth and selenium accumulation of grape, and screen out the optimal melatonin concentration that could promote grape growth and selenium (Se) accumulation.
      Methods  The cuttings of 'Xiahei' grape was used as the material, and the pot experiment was conducted to study the effects of melatonin on the growth, physiology and Se accumulation of grape seedlings under Se stress.
      Results  Different concentrations of melatonin increased the biomass and root/shoot ratio of grape seedlings to a certain extent, and melatonin with a concentration of 150 μmol L−1 had the greatest increase (18.22%) in the shoot biomass of grape seedlings. Melatonin treatment also increased the contents of chlorophyll and carotenoids in grape seedlings to a certain extent, improved the photosynthetic capacity and antioxidant enzyme activities, and decreased the proline content, soluble protein content, relative conductivity, and malondialdehyde content. Different concentrations of melatonin increased the Se content in grape seedlings, among which melatonin at a concentration of 150 μmol L−1 increased the Se content in the shoots of grape seedlings the most (112.71%). Regression analysis showed that there were quadratic polynomial or linear regression relationships between melatonin concentration and root biomass, shoot biomass, Se content in roots, or Se content in shoots of grape seedlings.
      Conclusion  Exogenous melatonin can promote the grape growth and Se accumulation under Se stress, and the concentration of 150 μmol L−1 is the best.
